TCVN 7220-1: 2002 do Tiểu ban kĩ thuật Tiêu chuẩn TCVN / TC 147/ SC1 "Phương pháp sinh học" biên soạn, Tổng cục Tiêu chuẩn Đo lường Chất lượng đề nghị, Bộ Khoa học và Công nghệ ban hành.

Giới thiệu

Chất lượng nước của một thuỷ vực có ảnh hưởng nhiều đến quần thể động, thực vật sống trong đó.

Phương pháp sinh học đánh giá chất lượng nước của các thuỷ vực đang được ứng dụng rộng rãi ở nhiều nước trên thế giới. Tổ chức Tiêu chuẩn hoá Quốc tế (ISO) đã ban hành một số tiêu chuẩn quốc tế về phương pháp dùng động vật không xương sống ở đáy cỡ lớn (Benthic macro-invertebrates) làm dữ liệu sinh học để phân loại chất lượng sinh học sông và qua đó để đánh giá tác động nhân tạo đến chất lượng nước sông. Các tiêu chuẩn quốc tế này đã được chấp nhận thành các tiêu chuẩn Việt Nam, như TCVN 6966-1:2001 (ISO 8689-1), TCVN 6966-2:2001 (ISO 8689-2), TCVN 7176:2002 (ISO 7828), TCVN 7177:2002 (ISO 8265), v.v.

Phương pháp sử dụng động vật không xương sống ở đáy cỡ trung bình (Benthic meio-invertebrates) để đánh giá chất lượng sinh học sông và qua đó đánh giá chất lượng nước sông sẽ được Tiểu ban Phương pháp sinh học thuộc Ban kỹ thuật Tiêu chuẩn TCVN/TC 147 Chất lượng Nước biên soạn thành một bộ TCVN trong thời gian tới. Tiêu chuẩn đầu tiên của Bộ tiêu chuẩn này đề cập đến phương pháp sử dụng Giun tròn (Nematoda) làm chỉ thị. Các tiêu chuẩn được biên soạn tiếp sau sẽ đề cập đến các đại diện điển hình khác của meiobenthos.

Cảnh báo: Khi làm việc dưới nước ở chỗ nước sâu và chảy, đặc biệt là nơi có nền đáy không ổn định, phải tuân thủ nghiêm ngặt các yêu cầu về an toàn trong khi lấy mẫu hiện trường.

1 Phạm vi áp dụng

Tiêu chuẩn này qui định và hướng dẫn sử dụng các dụng cụ và cách thức lấy mẫu định lượng giun tròn và các nhóm động vật không xương sống ở đáy khác cỡ trung bình (từ đây viết tắt là ĐVĐTB) bằng dụng cụ lấy mẫu hình vuông, hình trụ ở những nơi có độ sâu nhỏ hơn 500 mm (trong những điều kiện cụ thể, có thể áp dụng cả ở độ sâu của nước tới 1 m) và bằng gầu Ponar hoặc Petersen đối với các vùng nước sâu hơn 1 mét có nền đáy phức tạp: cát, sạn, bùn, đất sét, v.v.

Phương pháp này được áp dụng để lấy mẫu ở hầu hết các loại thuỷ vực như: sông, các dòng chảy và cửa sông [35]. Kết quả lấy mẫu bằng dụng cụ lấy mẫu hình trụ và hình vuông có thể cung cấp chính xác các dữ liệu định lượng về hiện trạng, tính đa dạng và sự phong phú về mối quan hệ của các đơn vị phân loại trong quần xã giun tròn và các nhóm ĐVĐTB khác.

2 Tiêu chuẩn viện dẫn

TCVN 5993: 1995 (ISO 5667-3: 1985) Chất lượng nước - Lấy mẫu - Hướng dẫn bảo quản mẫu và xử lý mẫu.

3 Thuật ngữ và định nghĩa

Trong tiêu chuẩn này, sử dụng các thuật ngữ sau:

3.1 Trầm tích đáy (benthic deposit)

Là các chất lắng tích tụ trên đáy sông, suối, hồ ao hoặc biển có thể chứa các chất hữu cơ được sinh ra do các nguyên nhân như xói mòn tự nhiên, các quá trình sinh học hoặc xả nước thải và cũng là nơi sống của động vật đáy.

3.2 Động vật không xương sống ở đáy (ĐVĐ) (benthic invertebrates)

Là nhóm động vật không xương sống, tồn tại và phát triển ở nền đáy của các hệ sinh thái thuỷ vực khác nhau như ao, hồ, đầm, sông suối và biển.

3.3 Động vật không xương sống ở đáy cỡ trung bình (ĐVĐTB) (benthic meio-invertebrates)

Là nhóm động vật không xương sống ở đáy có kích thước trung bình (có chiều dài cơ thể từ 42 μm đến 1000 μm), không nhìn thấy được bằng mắt thường và chỉ quan sát được bằng các loại kính phóng đại.

3.4 Chỉ số sinh học (biological index)

Là chỉ số thể hiện sự phong phú, mức độ tương đồng, mức độ đa dạng, tỷ lệ giữa nhóm loài (cá thể) chỉ thị trên tổng số họ (cá thể), ưu thế của các loài tại từng điểm nghiên cứu của quần xã không xương sống ở đáy cỡ lớn và quần xã không xương sống ở đáy cỡ trung bình.

4 Nguyên tắc

Lấy mẫu giun tròn và ĐVĐTB ở những vùng nước chảy, không sâu bằng dụng cụ lấy mẫu cầm tay nêu trong tiêu chuẩn này là tách và lấy một phần trầm tích đáy của nơi được lấy mẫu.

5 Dụng cụ lấy mẫu

5.1 Dụng cụ lấy mẫu hình vuông

Hầu hết động vật không xương sống ở đáy cỡ trung bình, trong đó có giun tròn thường sống trong lớp bùn dày từ 1 cm đến 15 cm cách bề mặt của đáy. Diện tích bề mặt đáy khoảng 20 cm2 là tương đối đủ để có thể thu được hầu hết đại diện của chúng. Dụng cụ lấy mẫu là thuổng kim loại hình vuông với cạnh 5 cm x 5 cm; thân và cán hình tròn dài 450 mm, có nắp đậy phía trên khi thao tác lấy mẫu.

5.2 Dụng cụ lấy mẫu hình trụ

Dụng cụ lấy mẫu là một ống kim loại hình trụ với phần trên có tay cầm và nắp đậy khi thao tác (hình 1). Dụng cụ lấy mẫu hình trụ có cấu tạo như sau: Gờ phía trên được đậy bằng một nắp nhựa hay kim loại có tác dụng bịt kín trong quá trình thao tác lấy mẫu trầm tích đáy và nhấc lên khỏi đáy, còn gờ phía dưới  (đáy) có răng cưa để tiện thao tác khi nền đáy là sạn và nhiều bã thực vật, độ sâu của mỗi răng cưa là từ 3 mm đến 10 mm, khoảng cách giữa các răng là từ 5 mm đến 10 mm. Đường kính của dụng cụ hình trụ thường là 55 mm hoặc 100 mm, chiều dài của thân ống là 400 mm hoặc 500 mm. Tay cầm hình tròn dài khoảng 70 mm với đường kính từ 15 mm đến 20 mm.

5.3 Gàu Ponar hoặc Petersen

Dụng cụ lấy mẫu loại này được dùng để lấy mẫu ở các thuỷ vực có độ sâu hơn 1 mét. Gầu Ponar như mô tả trong hình 2 có nguyên lý cấu tạo, kết cấu và kích thước giống như gầu Petersen. Gầu Ponar chuẩn có kích thước miệng gầu rộng 230 mm, diện tích ngoạm mặt đáy là 0,053 m2 và nặng 13,7 kg.

6 Dụng cụ phân tích trong phòng thí nghiệm

Ngoài các dụng cụ chuyên dụng và thông dụng trong phòng thí nghiệm cần thiết cho phân tích ĐVĐTB và giun tròn, cần có các dụng cụ sau:

6.1 Kính hiển vi

Dùng kính hiển vi quang học hoặc kính hiển vi soi nổi, có độ phóng đại từ 400 lần đến 1000 lần.

6.1 Kính lúp

Dùng kính lúp có độ phóng đại từ 28 lần đến 140 lần.

7 Quy trình lấy mẫu

7.1 Chọn dụng cụ lấy mẫu

Tuỳ thuộc vào mục đích lấy mẫu định lượng và điều kiện địa hình nơi tiến hành nghiên cứu, người thao tác lấy mẫu có thể chọn một trong hai dụng cụ lấy mẫu là thuổng kim loại hình vuông hoặc hình trụ.

Việc lựa chọn dụng cụ lấy mẫu ống (thuổng) kim loại kiểu hình trụ hoặc hình vuông, hoặc gầu Ponar/Petersen là hoàn toàn tuỳ thuộc vào kỹ năng lấy mẫu, kinh nghiệm của mỗi người và điều kiện tại chỗ. ở nơi nước chảy chậm hoặc nước sâu và có thảm thực vật, dụng cụ lấy mẫu hình trụ có nhiều ưu thế do ít phụ thuộc vào vận tốc dòng nước mà vẫn phát huy tối đa công dụng của nó. Ngoài ra, dụng cụ lấy mẫu kim loại hình trụ thường được dùng trong trường hợp khi nền đáy là bùn nhuyễn, cát mịn hoặc đất sét và đất thịt.

7.2 Dùng thuổng kim loại hình vuông

Để có các số liệu định lượng, thường thu trầm tích của 4 thuổng thu hình vuông trong một lần thu tại một điểm lấy mẫu. Dụng cụ lấy mẫu là thuổng kim loại hình vuông với cạnh 5 cm x 5 cm, thao tác nhẹ nhàng để không làm động mạnh tới các vùng bên cạnh, ấn nhẹ thuổng lấy mẫu sâu vào nền đáy 15 cm, đậy nắp phía trên của thuổng, rút lên và cho mẫu vào lọ nhựa dung tích 100 ml sau đó sẽ lọc qua rây 0,3 mm để loại rác và các mẫu đất, đá còn bám lại trên rây. Nếu đáy là cát thô hoặc cát mịn thì sẽ cho cát vào nước sạch, khuấy tròn mẫu trong lọ nhựa dung tích 1000 ml, sau đó gạn phần nước phía trên vào lọ đựng mẫu. Dùng đĩa Petri đổ từng phần mẫu lên đĩa, cho thêm nước lã sạch, khuấy tròn; quá trình này nhắc lại từ 7 đến 10 lần là đủ để tách tất cả động vật đáy không xương sống khỏi cát. Loại bớt nước trong lọ đựng mẫu và định hình bằng formalin 4 %. Công việc phân tích được tiến hành dưới kính hiển vi trong phòng thí nghiệm [35]. Đối với tách lọc giun tròn, ly tâm, phân tích và định loại, tiến hành theo quy trình nêu ở phụ lục A [32].

7.3 Dùng ống lấy mẫu kim loại hình trụ

Do hầu hết quần xã giun tròn và các ĐVĐTB khác như: Copepoda, Turbellaria, Ostracoda, Gastrotricha, Cnidaria v.v... thường sống trong lớp bùn dày từ 1 cm đến 15 cm cách bề mặt của đáy nên thực tế khi lấy mẫu chỉ cần diện tích bề mặt đáy từ 23,7 cm2 đến 78,5 cm2 là đủ để có thể thu được hầu hết đại diện của chúng. Để có các số liệu định lượng, thường thu trầm tích của 4 ống thu hình trụ trong 1 lần thu tại 1 điểm lấy mẫu. Thao tác cần nhẹ nhàng để không làm động mạnh tới các vùng bên cạnh, ấn nhẹ ống lấy mẫu sâu vào nền đáy 15 cm, đậy nắp phía trên của ống lấy mẫu, rút lên và cho mẫu vào lọ nhựa sau đó sẽ lọc qua rây 0,3 mm để loại rác và các mẫu đất, đá. Nếu đáy là cát thì cho cát vào nước sạch, khuấy tròn mẫu trong lọ nhựa dung tích 1000 ml, sau đó gạn phần nước phía trên vào lọ đựng mẫu 500 ml. Dùng đĩa Petri đổ từng phần mẫu lên đĩa, cho thêm nước lã sạch, khuấy tròn; quá trình khuấy nhắc lại 7 đến 10 lần là đủ để tách tất cả động vật đáy không xương sống khỏi cát và bùn. Loại bớt nước trong lọ đựng mẫu và định hình bằng formalin 4 %, công việc giám định được tiến hành dưới kính hiển vi trong phòng thí nghiệm [35]. Đối với tách lọc giun tròn, ly tâm, phân tích và định loại tham khảo cách tiến hành theo quy trình nêu ở phụ lục A [32].

7.4 Dùng gầu lấy mẫu Ponar hoặc Petersen

Đối với vùng nước sâu hơn 1 mét có cấu trúc nền đáy là cát, bùn, đất sét, đất thô, sạn v.v... thì việc sử dụng gầu Ponar/Petersen là biện pháp an toàn và hiệu quả nhất cho các mẫu định lượng. Để có các số liệu định lượng thường thu trầm tích của 4 gầu Ponar/ Petersen với tổng diện tích đáy là 0,1 m2 nền. Trầm tích được lọc qua lưới 0,3 mm để bỏ bớt rác, vụn cây và sỏi, được rửa trong rây có đường kính 400 mm, với mắt lưới 0,1 mm cho đến khi sạch. Sau đó mẫu ĐVĐTB được gạn lọc bớt nước và được lưu giữ trong lọ đựng mẫu dung tích 500 mm và được cố định bằng formalin 4 % [35]. Đối với riêng giun tròn (Nematoda) thì trầm tích không qua quá trình rửa bỏ rác và sạn trong các rây mà được cho vào lọ nhựa 500 ml và cố định bằng formalin 10 %. Các bước nghiên cứu, phân tích tiếp theo được tiến hành trong phòng thí nghiệm.

Chú thích - Tất cả các mẫu ĐVĐTB và giun tròn đã thu được, cần bảo quản theo TCVN 5993:1995.

TCVN 7220 – 1: 2002 is prepared by Technical committee TCVN / TC 147/ SC1 " Biological method", submitted by the Directorate for Standards and Quality (STAMEQ) and approved by Ministry of Science and Technology (MOST).

This English version of Vietnam standard gives the equivalent items and meanings in English language.

However, only the items and its meanings in Vietnamese language can be considered as Vietnam standard.

Introduction

Water quality of every waterbody has impact on plants and animal populations. Biological method has been widely used in many countries to assess water quality. Recently. the International Organization for Standardization (ISO) has issued standards on biological methods using macro-invertebrates as biological quality data for classification of rivers and assessment of human impact to water quality of rivers. A few years ago, these International Standards were adopted as Vietnam Standards such as TCVN 6966-1: 2001 (ISO 8689-1), TCVN 6966-2: 2001 (ISO 8689-2), TCVN 7176: 2002 (ISO 7828), TCVN 7177: 2002 (ISO 8265). Standards on methods of using meio-benthos for classification of rivers and for assessment of the biological quality of rivers have been drafting recently by TCVN Sub- Technical Committee: TCVN/TC147/SC1 “Water quality - Biological methods for water quality assessment”. That standards will be issued in serial by Vietnam national standards body in a near future.

This first of serial standards on the water quality assessment by use of bio-index, the TCVN 7220-1: 2002 mentions only to a method of using Nematodes as bio-indicators. Other typical benthic invertebrates with a view of biological indicators will be scope and subject metter for oncoming of standard development.

WATER QUALITY - WATER QUALITY ASSESSMENT BY USING OF BIOLOGICAL INDEX - PART 1: METHOD OF USING QUANTITATIVE SAMPLERS FOR NEMATODES AND MEIO-BENTHOS ON SUBSTRATA IN SHALLOW FRESHWATERS

Safety precautions - Working alone in water is not recommended in view of the risks from high current velocities, deep waters and unstable beds, safety requirements must be strictly obeyed during sampling.

1. Scope

This part of Vietnam standard gives guidance on the equipment and procedure for quantitative sampling of nematodes and other meio-benthos by quadrat samplers and cylinder samplers in water of depth less than 500 mm (although methods are described to permit sampling, under certain conditions, in a water depth of up to 1 m) and by Ponar/Petersen crab in watershed depth of more than 1 m with mixed sediments such as sand, graves, mud, clay etc.

This procedure is applicable to the sampling of all accessible aquatic habitats in river, running currents and estuaries. The results of using the above mentioned samplers provide precise quantitative data on the presence, diversity and relative abundance of taxa in nematode and other meio-benthos communities.

2. Normative reference

The following normative document contain provisions which, through reference in this text, contitute provisions of this Vietnam standard

TCVN 5593: 1995 (ISO 5667 - 3: 1985)    Water quality - Sampling – Part 3: Guidance on the preservation and handling of samples.

3. Interpretation

For purpose of this standard, the following terms apply

3.1. Bed deposits (Sediment)

All deposit matters in beds of rivers, watercourses, streams and estuaries that may contain organic substances which had been resulted from natural erosion and biological process or discharge of waste water. Sediment is a habitat for all meio-benthos and nematodes.

3.2. Benthic invertebrates

Group of benthic animals using sediment as a dwelling at the bottom of aquatic environment

3.3. Meiobenthos

All benthic invertebrates that are not easily visible without optic magnification and usually have the length from 45 μm to 1 mm.

3.4. Bio-Index

A biological index designed to evaluate the abundance, similarity, diversity and proportion level between each group individuals in terms of the total families at every investigating sites of meio- benthos communities.

4. Principle

Sampling of nematodes and meio-benthos in shallow and running water by collection using manual cylinder and quadrat samplers as mentioned in this standard is isolation of a portion of sediment at sampling sites in the water body.

5. Sampling equipment

Depending on the sampling quantitative purpose and landscape conditions of the investigating areas, one of the two type of sampling corers can be selected, which is a cylinder sampler or a quadrat sampler.

5.1. Quadrat sampler

In general, allmost meio-benthos living in 1-15 cm depth of the bottom of an aquatic environment. With a surface area of about 20 cm2 sampled by quadrat sampler, it is possible to collect most of meio- benthos. Quadrat sampler used for sampling is a square of dimensions 5 cm x 5 cm with the cylindrical handle of 450 mm length and closing cap on the opposite.

5.2 Cylinder sampler

The sampler consists of an open-ended cylinder with handle and closing cap at the upper end (Figure

1). The upper edge is covered by a plastic or metallic cap with a view to closing the cylinder when the sediment sample is collected and lifted up from the river bed. The lower edge serrated teeth for the user's convenience when sampling sites are with much gravel and debris of plants. The teeth are 3 mm -10 mm deep and separated at the distance of 5mm -10 mm. The cylinder is 400 mm - 500 mm long and has a diameter of 55 mm or 100 mm, designed with a round handle of 70 mm length and diameter of 15 mm - 20 mm.

5.3 The Ponar or Petersen grab

The Ponar grab is used to sample in watershed of more than 1 metre depth. It is similar to the Petersen grab in size, structure, weight as described in figure 2. A standard Ponar grab, weighs 13,7 kg with the mouth opening in 230 mm width, which can collect a sampling area of 0,053 m2 on river bed. This sampler is best use for substrate with sand, gravel or small rocks.

6. Analyzing equipment in laboratory

Apart from specialized laboratory equipment, for meio-benthos and nematode analysis the following tools are needed:

6.1. Microscope

The optical microscope or stereomicroscopes with magnification from 400 to 1000.

6.2. Binocular stereoscope/ magnifier

The stereoscope with a magnification from 28 to 140.

7. Sampling procedure

7.1. Choice of sampler

The choice between the cylinder type sampler and quadrat type sampler or Ponar grab depends on personal preference based on operating experience and the prevailing conditions. In slow-flowing water, or in deep waters and among vegetation, the cylinder sampler has the advantage of being less dependent on velocity of flow to operate successfully. Besides, when in the substrate is presence mud, sand, clay and loam, the metallic cylinder sampler to be preferred to use

7.2. Quadrat sampler

In order to collect quantitative samples of the sediment, at every investigating site the sampling are operated gently with 4 square corers to avoid disturbing the surroundings. The square sampler which has the edges of 5 cm x 5 cm is pressed on sediment to depth of 15 cm, then the upper opening cover is closed. The sediment in corer is picked up from the bottom and put to the plastic bottle of 100 ml and washed through a net sieve with the mesh size of 0,3 mm to remove silty materials. In case of sediment is sandy, it is convenient to place the samples with sandy particles in a bottle of 1000 ml. Pour small part of the sample in a Petri disk, then add pure water and carefully stir to dislodge meio- benthos from sands. Immediately after stirring, the upper layer of water is poured into a bottle (100 ml) and this washing process is continued and repeated for 7-10 times, which relatively all meio-benthos is released from sand particles. The bottled samples are fixed with 4% formalin and analyzed in laboratory by using microscopes. For the centrifugation extraction, analysis and identification of nematodes, the procedure is followed in accordance with the procedure described in Annex A of this standard.

7.3. Cylinder sampler

Due to most of meio-benthos such as Copepoda, Turbellaria, Ostracoda, Gastrotricha, Cnidaria etc. reside bottom surface in a sediment layer of 1-15 cm depth, the bottom area of 23,7 cm2 to 78,5 cm2 to be collected is actually sufficient for obtaining samples with predominant species of meio-benthos. The cylinder sampler is pressured into sediment to the depth of 15 cm, then the upper opening cover is closed. The sediment in corer is pick up from bottom and put to the plastic bottle of 1000 ml and washed through a sieve net with the mesh size of 0,3 mm to remove silty material from the sample. In case of sediment is sandy, place the samples with sandy particles in a bottle of 500 ml, pour each part of the sample into Petri disks and add pure water and carefully stir to dislodge meio-benthos from sands. Immediately after stirring, the upper layer of water is poured into a bottle (100 ml) and this washing process is repeated for 7-10 times, which almost of meio-benthos released from the sand particles. The bottled samples are fixed with 4 % formalin and analyzed in laboratory by using microscopes. For the extraction centrifugation, analyses and identification of nematode, the procedure is carried out in accordance with procedure described in Annex A of this standard.

7.4. Ponar or Petersen grab

For the watershed of more than 1 metre depth with the bottom of sand, mud, clay, loam, rock, etc. It is the most effective and safe method to use Ponar/ Petersen grabs to collect quantitative samples.

For sampling quantitative data of sediment, at every investigating sit the Ponar/Petersen grab may be used to conduct 4 samples of sediment so that the grab collects fully 0,1 m2 from the bottom surface of the site. The sediment in grabs is filtered through a sieve net, with the mesh size of 0,3 mm to remove silty materials and washed through a net with frame 400 mm in diameter and the mesh of 0,1 mm. Afterwards, the samples of meio-benthos are decanted and stored in the bottle of 500 ml and fixed in formalin 4%. As per nematode samples, the sediment will be put into plastic bottle of 500 ml and fixed in hot formalin 10 % without being washed and extracted in sieves. The next steps for research and analysis will be performed in laboratories.

Note: All collected samples of meio-benthos and nematodes should be stored in accordance with the TCVN 5993 – 1995.
